- A phase II study involving adult female patients, >=18 years, with stage IIIB or IV HER2-positive (by IHC and/or in situ fluorescence) metastatic breast cancer. All patients were previously treated with trastuzumab and a median of three chemotherapy lines. 41 patients received 50 mg afatinib once-daily until disease progression and the primary endpoint was objective response rate. 4 patients (10% of 41 treated; 11% of evaluable patients) had partial response and 15 patients (37% of 41) had stable disease as best response. Thus a total of 19 (46% of 41) achieved clinical benefit. Median progression-free survival was 15.1 weeks.
