Description
20 patient-derived colorectal cancer xenografts were treated with cabozantinib, a c-MET and VEGFR2 inhibitor. 16 xenografts showed sensitivity (as defined by TGII: tumor growth of treated compared to control ≤ 20%). Among the genes tested for mutations (KRAS, TP53, PIK3CA, MET, VEGFR2, RET and AXL), PIK3CA mutations (4 PDTX) showed a significant enrichment in sensitive xenografts. PIK3CA mutations were observed in 4/5 of the most responsive tumors. Treating xenografts derived from isogenic PIK3CA wildtype or mutant cell lines corroborated this finding with PIK3CA mutant xenografts showing greater sensitivity to cabozantinib.
