- This was a retrospective clinical study of 168 metastatic colorectal cancer patients who received first line chemotherapy (made up of 5-fluorouracil (5-FU) only, 5-FU + oxaliplatin, 5-FU + irinotecan, or 5-FU + oxaliplatin + irinotecan) alone or with monoclonal antibodies (bevacizumab or cetuximab). It assessed the relationship between PIK3CA mutation status in primary tumors and post treatment PFS. Of 168 patients, 142 had wildtype PIK3CA and 26 had mutations in PIK3CA on either exon 9 or 20 (exon 9 mutations: E542K, E545K, E545G, E545D, Q546K, D549N. Exon 20 mutations: R1023Q, H1047R, H1047L, G1049A). The study found that patients harboring a PIK3CA mutation did not have a significantly worse outcome than patients expressing wildtype PIK3CA, regardless of first line chemotherapy regimen or use of monoclonal antibody treatment.
