Description
In this multicenter phase I study of abemaciclib, a total of 225 patients were enrolled: 33 in dose escalation and 192 in tumor-specific cohorts. Radiographic responses were achieved in previously treated patients with breast cancer, NSCLC, and melanoma. The disease control rate for the KRAS-mutant population was 55% (16/29) compared to 39% (13/33) in KRAS wildtype cohort. Stable disease lasting ≥24 weeks was achieved for 31% (9/29) with KRAS-mutant disease vs 12% (4/33) with KRAS wild-type disease. Median PFS was 2.8 months and 1.9 months for the KRAS-mutant and KRAS wild-type populations, respectively.
