Description
In an in vitro study, a Ba/F3-p210BCR-ABL1 cell line was ENU-mutated and exposed to graded concentrations of imatinib, nilotinib or dasatinib to identify resistance mutations. The L384M mutation in the ABL1 portion of the BCR-ABL1 fusion protein was associated with resistance in 50nM nilotinib (1 clone recovered, ~2% of viable clones in this treatment) and 2 μM imatinib (1 clone, ~0.7%). L384M was one of 3 novel mutations that were not identified in imatinib resistant patients from a review of 20 publications.
